Real were expected to extend their 22-match winning streak in all competitions with a victory at Mestalla and they took the lead thanks to a penalty from Cristiano Ronaldo.
However, second-half goals from Antonio Barragan and Nicolas Otamendi earned Valencia a surprise win.
"We played a good match and didn't deserve to lose," the Italian told reporters after the match. "But, in football, anything can happen.
It's a defeat which we eventually had to suffer. Congratulations to Valencia — they fought hard. But we worked well and we created chances. There is nothing else to say."
