Rane has forwarded his resignation to the Maharashtra Chief Minister Prithviraj Chavan, sources said.
Nilesh lost to Shiv Sena nominee Vinayak Raut in a close contest.
When contacted, Maharashtra Pradesh Congress Committee (MPCC) president Manikrao Thakre confirmed Rane's resignation.
"Although Rane has tendered his resignation over the loss of his son Nilesh, the same will not be accepted. We will hold discussions with him and find a way out," Thakre told PTI.
Meanwhile, Shiv Sena president Uddhav Thackeray has left for Ratnagiri district where he is likely to address a 'victory rally' for Raut.
