Samantha Ruth Prabhu made a sparkling debut with 'Ye Maaya Chesave' in Telugu which was a hit. She was starred opposite Naga Chaitanya in this sweet love story. Samantha played Jessie in this complicated inter-religion love story. Her performance was praised by critics and audience alike and was said to be 'the one to watch out for'

In 2012, Samantha starred in SS Rajamouli's fantasy film 'Eega'. The film's narrative is in the form of a bedtime story told by a father to his daughter. Its protagonist Nani, who is in love with his neighbour Bindu, is murdered by a wealthy industrialist named Sudeep, who is attracted to Bindu and considers Nani a rival. Nani reincarnates as a housefly and tries to avenge his death and protect Bindu from an obsessive Sudeep.

Samantha played the role of Bindu and received the Best Actress award at Filmfare

Samantha plays Yamuna in the 2015 Tamil film 'Thanga Magan' opposite Dhanush. The two actors play a married couple who try to uncover the reason behind Dhanush's character Tamizh's father's suicide

Directed by Atlee, Samantha starred as Vijay's wife in Theri. She is seen in the flashback segment of the film where the events to her murder is unfolded and how she saves her daughter and makes her police officer husband vow to always protect their daughter

In the 2017 film 'Mersal', directed by Atlee, Samantha was reunited with her Theri co-star Vijay. She plays a doctor in the film and fiance of Vijay's character Maaran. Vijay is seen in a triple role in the film

In 2018, Samantha transformed herself completely into village girl Ramalakshmi in Sukumar's film 'Rangasthalam. “Everything about the film and my role is on a different tangent. Sukumar was very clear that he didn’t want to see Samantha at all. I had to walk, talk, and even laugh like Ramalakshmi. There was a scene where I laughed like I normally do, and Sukumar asked for a retake and asked me to make the laugh sound ugly," she had once said
